TNFR2

signaling-pathways

TNFR2 is the second of the two receptors for tumor necrosis factor. TNF signaling is a central immune axis, but its downstream output is strongly receptor- and context-dependent, and TNFR2 is associated with immunoregulatory rather than straightforwardly pro-inflammatory programs.

Reported features of TNFR2-mediated signaling include engagement of PI3K-associated immunoregulatory programs, suppression of NF-kappa-B transcriptional activity, and promotion of C/EBP-beta-linked macrophage polarization. This makes TNFR2 mechanistically distinct from TNFR1 and a candidate target for shifting macrophages away from immunosuppressive states.
