Decitabine is a clinically approved pyrimidine nucleoside analog that acts as a DNA hypomethylating (demethylating) agent, incorporating into DNA during replication and inhibiting DNA methyltransferases. It is used to reverse epigenetic silencing of genes inactivated by promoter hypermethylation, including tumor-suppressor genes silenced in cancer.
